Dear members,
Recently I isolated several fungi from soil but the identification is
really hard ---- the morphological method is too sophisticated to a novice
like me. I notice that ribosomal DNA sequencing based method is widely-used
in papers, which is relative simple and convenient. However, the primers
used by different groups seem to be diverse, which may target either 18S
ribosomal or ITS sequences. I try to looking for some reviews or articles,
but which seems to be rare. So, may anybody can tell me which primers
should be choosed, and how about the reliability of phylogenetic
relationship speculated from DNA sequence?
Thanks very much in advanced!
