We are trying to retrogradely label motor neurons in day 13 mouse embryos using fluourescent(sp?) beads but are having a great deal of trouble getting the embrynic tissue to survive for 24 hours since the embryos are dissected out of the mother. Can anyone offer any suggestions for what has worked for them(i.e. media used, drugs for increasing retrograde transport rate...). We need to increase the viability of the tissue enough to plate the spinal cord neurons the following day.
Thanx in advance for any help that is provided. =)
