I'm trying to do a whole cell patch clamp study on smooth muscle cells
isolated from rat middle cerebral artery. However the cells when
isolated, have a very inelastic, noncompliant membrane, and I have been
having trouble getting good seals. Can anybody suggest a reason for this
problem (solution osmolarity ?) ?
