succinate dehydrogenase

Dear Folks,

We are interested in measuring succinate dehydrogenase activity in crude
extracts of seedlings without first purifying mitochondria.  As yet, our
assays of crude extract are not giving us any activity above background
(without succinate present).  It occurred to me that perhaps the assay isn't
sensitive enough to measure activity in crude preps.  Does anyone have
experience with succinate dehydrogenase assays?  Must one purify mitos to
measure activity?
