respiratory control in mitochondria

Hi Plant-eders,

We isolated cauliflower mitochondria today in Plant Physiology lab
following the protocol in Carol Reiss' Experiments in Plant Physiology
manual. The mitos took up oxygen, and were inhibited by malonate (when
fed succinate) and cyanide. What they didn't do was show any trace of
ADP limited O2 uptake suggesting that they were completely uncoupled. I
used fatty acid free BSA in all of the isolation and washing media.
Does anyone have experience with this problem and how to solve it?
