I would think the best approach in any system is to express it from an
inducible promoter with a very low constitutive level. This is easiest
in bacteria, but you can od it in yeast etc. too. The same would apply
in plants. If it messes up membranes, then you have to go the inducible
route.
As for choice of plant - go with leaf expression in tobacco, you would
get masses of easily extracted tissue. Maybe you could export it from
the cells into the intracellular space, or the culture medium in the case
of yeast, bacteria or animal cells.
