Applying Hydric stress to wheat plantlets

Good Morning,
I'm trying to apply dehydration to 7 days old wheat plantlets. In the litterature, the most used 
protocols talk about applying a 85% relative humidity in a dessicator (with 25% glycerol solution).  
Some authors aplly depression (-12 mPa) and some others not. And I don't know how should i 
do?
