from your post I did not clearly get what your problem was. If you want to check whether
your expressed protein has the cysteines you expect (or whether some of them are
involved in disulfide bond formation) you could use Ellmanns assay (DTNB) to quantitate
free Cys (the protein has to be denatured). The Thannhaeuser assay (reduction with
sulfite) allows determination of all cys, difference gives disulfide bonds.
