MICROSEQUENCING N-BLOCKED PROTEINS can be done more quickly and efficiently
than traditional methods involving enzymatic cleavage/HPLC separation of
peptides. We have developed a method that can identify an N-blocked protein
using a single sequencer run and avoids the problems inherent in the
traditional HPLC recovery of peptides (e.g. over-representation of
contaminant peptides, inefficiency of enzyme cleavage on sub-microgram
amounts of protein, and aggregation of peptides from very hydrophobic
