Getting a good thrombin, as the others have stated, is key to success.
In our lab, we have found that the best and most specific thrombin is
the human isolate offered by Sigma (T-6759). We have performed
incubations at RT of up to 30h without significant secondary degradation
(in cleavages of 1.5kD peptide from GST).
