8M urea and 6M Gu-HCl: can they lysis yeast cell wall

Think about an additional mechanical means to disrupt such as a bead
beater, sonication, pressure disruption. I do not believe those agents
break the cell wall. Surely there are well worked out protocols for yeast
protein extraction.
