[Protein-analysis] Batch vs column chromatography

Hello,

I'm puryfing a protein using Ni-NTA (Qiagen) technology. Recently I
load the crude extract from my protein onto the column, but I'm
considerating using of batch method. Tell me please if it is reasonable
to add about 1mL of resign to the 150mL of crude extract and leave it
vigorously shaking overnight? Are these proportions of volumes and
times are reasonable?
