[Protein-analysis] troubleshooting for 2-D

Hi all,

I have a problem with my 2-D. My 2-D mostly shows stacking bands like
first dimension gel. Even if there are spots, apart from spots vertical
bands are also present.

I would be obliged if anyone can guide in a proper direction for 2-D.
