[Protein-analysis] Ask

Hello ,
I have problem with protein expression in cos-7 and
HEK293 cell , I can not get reasonable amount of
protein , I have used control one and the result was
huge amounts of protein while the others not .I want
to tell you more , first I have mutated some amino
acids , I have tranformed into Ecoli Top10 ,thereafter
I transfected into cos-7 by using a reagent lipofectin
. I could not get reasonable amount even I scaled up ,
when I have purified my protein by affinity
chromatograghy in copper column , I could not see any
band at all .
