One of the major problems in structure determination is the
determination of the structures of membrane proteins. Success
has been achieved where the soluble portion can be cleaved off and
the structure of that portion determined. The structures
of true membrane proteins are few and far between. The major reason is
the difficulty in obtaining diffraction quality crystals. Some success
has been reported with two dimensional crystals for use
in electron diffraction.
