Membrane proteins in dihybrid?

We had good luck using the two-hybrid system to detect interactions among
proteins associated with the inner mitochondrial membrane. One of the
proteins, PET122, doesn't have an obvious membrane spanning domain but
behaves like an integral membrane protein (when overproduced). Another,
PET494, also appears to be tightly associated with the membrane, but PET494
only worked when a putative membrane spanning domain was deleted. The
third, PET54, is soluble and peripherally associated with the membrane.
