RE: yeast colony PCR.  

There was a long discussion on this group a while back.  You might try 
searching the yeast section of the stanford gopher server to find it.

We found that preboiling was death, and got our best results when we used 
fresh colonies (2 days old), put them straight in the tube with all the 
goodies, put the tube in the machine and turned it on.  This was with 
saccharomyces.
