Danny,
Check the molbio.methods-reagents archives for more detailed 
correspondence, but the simple way is to grow a fresh plate of your yeast 
and pick a small colony with your toothpick. Swizzle (scientific term) in 
your PCR mix and go. I use hot start, which greatly improves specificity.
