PET56 clone

Dear yeasties:

The original transplacement of his3delta200 did not confer a petite
phenotype (Fasullo and Davis, MCB 8: 4370-4380). However, sequence
analysis of the allele revealed that the deletion extends into the
pet56 promoter and that pet56 RNA levels are 20% of those observed
in WT strains (Struhl PNAS 82: 8419-8423).The deletion fuses poly
dA:dT sequences from the DED1 promoter to create a bidirectional 
promoter for both DED1 and PET56. Apparently, in some strain back-
grounds, spontaneous deletions of the stretch of polydA:dT can 
confer a higher frequency of petites, and a petite phenotype may
be linked to the His- phenotype.
