3-AT, YMM? YPD?

I don't know if it's best, but I use synthetic media, e.g. complete 
synthetic minus hisitidine etc. I autoclave the media first, then let it 
cool some before adding the appropriate amound of 3AT. I make up the 3AT 
in H2O as a 1 M stock and store it at -20. I have found that adding 35 
glucose to the media instead of my usual 2% increases plating efficiency 
on this media, but this is somewhat anecdotal. It is crucial to leave the 
histidine out of the media; thus, I would not expect rich media such as 
YEPD to work.
