[Q] yeast colony hybridazation

Dear yeast netters,

Would you teach me the protocols of yeast colony hybridazation?

I am trying yeast two hybrid screening against random-primed library.
I want to exclude the bait cDNA itself from positive clones as a result
of homophilic interaction. I took PCR into account, but I have no idea
how to disign the primer, because I havn't determine the interface domain
of the bait cDNA. 

I will be grad if you tell me other ideas than colony hybridazation.
