I'm about to isolate S.pombe yeast nuclei using the method of Aris and Blobel
(Methods in Enzymology vol.194), but I am looking for the best protocol to lyse
these nuclei in order to obtain a NATIVE protein extract - for use in gel-shift
assays, SouthWestern blot analysis etc.
I would be most greatful for any guidance on this subject.
