Greetings. Obviously reducing sugar assays indicate hydrolysis, but not
location. In my work, the only way of identifying location of glucanase
cleavage is by combining reducing sugar assays with a glucose oxidase test
or running the enzyme assay over a sugar separation column on a HPLC. I
routinely looked a residual sugars in fermentation broths using a Sugar
Pak column by Millipore. The only headache with sugar analysis by HPLC is
that it requires refractometry which is a pain.

I hope this is some help.
