Hi Dave! Have you tried to increase the salt concentration in your buffers? You might get very specific signals up to 300 mM NaCl? Another possibilty would be, using different fixation times. Sometimes, short fixation times increase the non-specific background. Another question: how did you affinity purify your antibodies?? Hope this helps! Anne
