Dear Olav

In my experience it is a question of getting the balance between a firm 
enough pellet (so you don't lose all the protoplasts), and the possibilty 
of breaking the protoplasts. I would say somewhere between 2000-3000g, 
for about 5 minutes is about right.
I used to find the protoplasts were quite hard to resuspend after 
treatment but be patient!
