Hi,
I don't know if this answers the question, but you may try the plasmid
shuffling experiment : your disrupted/mutated gene is on an episome
which you may counter-select (URA3...). This leads to a good growing
strain, and may be transformed as you like. Thereafter you may counter
select your first plasmid (5-FOA), and fish a complementing gene...
To resume : transform your slow growing yeast into a rapid growing
yeast and use the LiAc method, which seems to be the best one.
