Try the combined works of Guarente (start with Meth Enzymol 101:181
(1983)).  You can use chloroform and SDS to permeabilize the cells,
ONPG as your substrate, and normalize to culture OD.  The other 
classic method uses protein assays to normalize.  Maybe the 
Guthrie and Fink book or the red book also have protocols?
