3-AT Maximum?

Hello All, I have a question that has been plaguing me for while. In yeast
two-hybrid, using a Gal4 UAS::HIS3, "unwanted" bait activation is diminished
by using the HIS analog, 3-AT. Literature (Elledge specifically) says that
50mM 3-AT is the maximum one can use. Does anybody know why? I have heard
people using up to 100mM. Is this counter-productive?

Any response to this question is greatly appreciated.
