I am a graduate student trying to express a plant small heat shock protein
in yeast. I am looking for an expression vector with the following
1. tryptophan marker
2. yeast terminator/poly A site
3. Good multiple cloning site for my gene (EcoRI=ideal)
4. Nice strong promoter
I've checked most of the major companies (eg. Invitrogen, Stragene...) and
the ATCC. For a second experiment a HIS4 or URA3 marker would also work
(this would be similar to the Invitrogen pYES2, which can be purchased
for $220 if I REALLY need it. Has anyone tried pYES2? What do you think?).
Expression level is the bottom line, does higher copy number generally
give better results, all other things being equal?
