Jae, In S.cerevisiae, we have found that a 15 minute formaldehyde (3.7%) fixation on ice maintains the GFP signal when using the S656T GFP derivative. This may or may not work in pombe (haven't tried). Good luck! Mark Flory Dept. of Biochemistry University of Washington Seattle
