In reply to the query about removing cell walls to generate spheroplasts or
to lyse yeast cells, using either Zymolyase or other enzymes, the sensitivity
of the yeast cells to degradation depends strongly on the strain, density of
growth, and nutrients.  Thus, conditions are fairly reproducible if all
variables are held constant; if not, you must optimize your system, probably by
varying time and/or concentration of enzyme.  In general, it seems that log
phase cells (OD600 of 1 or less) are more sensitive to most enzymes than cells
later in growth.  Carbon source may also make a difference.
