Hello! Can anyone explain why my DNA isolated from strains on FOA plates won't cut with HindIII (despite repeated attempts at purification)? The identical strains grown on YEA or -ura plates give fine, cuttable DNA. The strains express low levels of ura, and thus grow about equally badly on FOA and ura, yielding similar ammounts of DNA. Any ideas?
