does you bait vector have a cycloheximide sensitivity gene on it? (CYH2). if it does, you can grow your yeast on -leucine media containing cycloheximide, which will result in the death of all clones harboring the vector containing the CYH2 gene. you should then have only the vector bearing your prey cDNA. hope this helps! don
