yeast nuclear extract

Hi,

I have two questions:

1. I'm going to prepare a nuclear protein extract from yeast cells which
then will be used for a DNA gel shift assay. Does somebody know a good
working protocol or some tricks or has some useful advice?

2. In this regard, how many micrograms of nuclear protein does one
obtain from one liter of yeast cell grown in exponential phase? What is
the ratio of mass of nuclear protein to total protein for yeast (in
percent)?
