I don't know what the protein yield should be, but I don't expect you'll
see it by total protein staining of lysates, if that's what you mean.  Can
you see it by blot?  Otherwise, a key factor is usually protease
inhibitors - make sure you're using a very robust cocktail to block
proteolysis.
