Basically take your yeast sample and add formaldehyde to about 1%.
Leave at 4C for a while. You can then spin them out and resuspend in
1ml 70% alcohol until you are ready to count. Then spin out the cells
and resuspend in 100ul of H2O to rehydrate and do your budding index.
