hi all

              Im trying to use the DAPI staining for budding yeast
saccharomyces cerevisae  BY4742 , but the dye is not entering the yeast
cells .
              Is it better  to prepare spheroplast  befroe incubation with
DAPI ? or DAPI is permeable to live yeast cells .


                please help me
