Andreea,
Sometimes you can increase sporulation efficiency by first streaking out
your diploid strain onto presporulation medium before putting it on KAc
plates. You can either order a mix or make it yourself - we use 4.8g yeast
extract, 1.8g peptone, 60g d-glucose, and 9 g agar for 600ml.
