[Yeast] Yeast spheroplast RT-PCR

Dear all,
 
Has anyone done RT-PCR from spheroplast?  I mean using spheroplast as template without extracting total RNA.  Originally I just used water to lyze spheroplast but all of a sudden my RTPCR has stopped working.  This is telling me I have been working on the limit.  Any idea how to eliminate the genomic DNA/cellular protein contaminant while keeping the integrity of mRNA?  Thanks!

BTW, RT=reverse transcription.
