Hi,
I have been doing a few Yeast 2-Hybrid screens with two baits both expressed at
high levels in EGY48 with pSH18-31. After a 40min heat shock I recovered in
YC-WUL 2%Galactose media for 6hours to induce the GAL1 promoter. However when
I tested my candidates for B-Gal activity I'm unable to get any color change
with my filter assay. I am using two +controls, a pFos/pJun in EGY48 and a
pFos/pJun in L40. The L40 control works well but the EGY48 is still white,
similar to all my candidate colonies. Does anyone have an advice on how I
could possibly get this B-Gal to work in EGY48?

So far I have only been able to screen these candidates by replica streaking
colonies on Galactose and Glucose plates to test the specificity of the Leucine
reporter but it would be nice to confirm these with both of the reporters.

On a side note I am also looking for another vector plasmid, pEG202-NLS or
something similar that uses a His selection. Origene only sells this as a part
of their yeast 2-hybrid kit and not alone. If anyone has an idea where I could
purchase this plasmid for a reasonable price it would be most appreciated.

Thanks,
Thomas
