Hi!
When you are plating your URA+ yeast cells on 5-FOA, the chance of getting false positives which are URA+ is very low. But you will get large number of aneuploid colonies (with unwanted mutations) which are URA-. So, it is recommended that whenever you are using this compound you should check your colonies with Southern hybridization, rather than PCR.

Hi,
I was wondering what false positive rate to expect when plating yeast on
5-FOA. Are these false positives due to Ura+ colonies that manage to live on
this media for stochastic reasons (and can be screened for by replica
plating onto 5-FOA a second time), or do they represent mutations that allow
survival of Ura+ colonies on 5-FOA?
Any help, even general comments on false positives and 5-FOA, would be
greatly appreciated.
