[Cell-biology] Floating Cell Clusters

I am working with primary muscle cells and C2C12
cells.

My question is:
Normally with cell cultures what causes cells to
cluster and float in the medium ?

Also is there a quick method of testing the viability
of the cell cluster (other than using Trypan Blue) ?
