We are embedded among several dynamic research groups actively working to understand the mechanisms of genome maintenance in complete molecular detail. Genome maintenance refers to the collective processes that preserve genetic information encoded in the chemical structure of DNA and faithfully pass this information on in cell division. The new projects are aimed at understanding the molecular interactions and structural dynamics that drive homologous recombination repair of DNA breaks. These processes are vital to cellular survival and important for cancer avoidance in humans. We approach this problem using cutting edge methods of dynamic structural biochemistry. We use scanning force microscopy to determine the molecular architecture of individual protein-DNA complexes with nanometer resolution. Dynamic interactions are defined in real time in active conditions using single-molecule fluorescent microscopy. Within this project the specific aims are: (1) to understand controlled assembly of active recombinase filaments on DNA in need of recombinational repair, (2) to reveal the molecular interactions that result in the products of recombination becoming the substrates for DNA synthesis, and (3) to define the structural changes required for homologous recombination reactions to occur on DNA packaged in nucleosomes. THE PROJECTS PhD student project is aimed at determining the requirements for forming functional filaments of Rad51 on DNA in need of recombinational repair. A filament of the human Rad51 recombinase bound to single stranded DNA is the catalytic core of the DNA strand exchange reactions of homologous recombination. In addition the influence of recombination mediators and specific DNA substrates on Rad51 filament dynamics will be determined. Post-doctoral projects have two broad goals, (1) to analyze the molecular events resulting in hand off of a recombinase created DNA joint molecule to a repair polymerase and (2) to analyze the mechanistic complexity of recombination reactions occurring in the context of nucleosomal packaged DNA. This work will involve defined in vitro reactions validated biochemically, analyzed by scanning force microscopy, and/or single complex fluorescent microscopy. There is considerable potential to develop this line of research in new directions including novel analysis techniques and methods.
