I tried to use the pTracer vector (pTracer-CMV) from Invitrogen in
both human fibroblasts and 3T3s. This vector contains a SV40 promoter that
controls GFP expression and a CMV promoter that drives the expression of
the gene of interest. My problem is that even with the empty vector (no
insert), GFP expression at 48-72h post-transfection is incredibly faint
(either on living or fixed cells, using FITC filter). Does anybody have any
experience with this vector ? Is it known not to work great ? Any
suggestions ?
