Hi everyone!
Is there any plasmid containing a signal peptide for ER synthesis of
the protein fused to its N-terminus? I would need it because : i would
like to make a series of deletions of a transmembrane protein fused to
the c -terminus of egfp. If i do so, the natural signal peptide of my
protein will become an internal part of the fusion and therefore
inactivated which is not desired! I know I could fuse it to the
n-terminus of Egfp, but this would be very complex because every
single mutant that i create had to be in frame with egfp to allow
fusion protein expression.Therefore i would prefer to have a signal
peptide in frame with egfp and fuse my protein without signal peptide
to c-terminus of egfp and create my deletion series from the very
c-terminus of the whole construct.
Any suggestions are highly appreciated!!!
