We are interested in loading the AM ester forms of Fura-2 or Indo-1 in the
yeast, Saccharomyces cerevisiae. We were not successful when we tried and
found in the literature that others could only load yeast with the
pentapotassium forms of Indo-1 or Fura-2 by electroporation or at very low pH.
Does anyone know the reason for this failure to load the AM ester forms of
these dyes? Do yeast lack the esterase to cleave the AM esters or are these
dyes impermeable to the yeast cell wall ?
