HELP! Why are my T cells normal by FACS?????

Make sure you are checking absolute cell numbers.  Also, consider checking
the percentages of B cells in the different groups.  If the B cells are
increasing in large amounts, the higher percentages that they generate on
FACS analysis may be overshadowing any changes you see in your other
populations.  For example, if you are staining for CD8CD44 and your
percentages are holding at 10% while your B cells are doubling, thus
falling into your double negative population for the most part, the
relative percentage of CD8CD44+ cells inside of the T cell population may
actually be increasing.  Also, consider the possibility that as the
populations become more active they may actually be leaving the node and
spleen to go where they are needed.  It's hard to tell based on the
information given and considering my unfamiliarity with your system, but
those are things that I have had to take in account with my research.
