We are trying to generate CTL from SIV and HIV-2 infected macaques. To
support macaque T cells in culture, we and others found that recombinant
human IL-2 did not work very well but that a conditioned medium (PHA sup
from human PBMC--a commercial product we got from Schiapparelli
Biosystems, Inc) was very good for growing cells. Unfortunately the
cells very quickly seem to develop LAK-like killing. Does anyone have
any experience with this? Specific questions:
1) Do macaque NK or LAK cells kill K562 (which is how we measure this)
2) Are there better defined cytokines for growing macaque T cells
3) Would it help to decrease the percentage of conditioned medium--we
use it at 10%
Any thoughts would be appreciated
