Dear Netters,
We are dealing with lymphocyte (t cell) cell cultures and are using media
prepeared with 10% serum, rest RPMI (as usual).
This media may be sufficient for short time cultures, but the question
arises whether they REALLY are sufficiant for long time cultures too. All
(necessary and not contained in RPMI) components are thinned tenfold,
maybe that NORMAL (not transformed) cells suffer damage by being depraved
from eg. hormons for a long time (and so arteficial effects are produced).
However, serum enriched media has suppressive (toxic) effects.
So, has anybody experience in prepearing serum enriched (maybe 50-70%),
non suppressive (toxic) media?
Of course, also reports of failure are welcome!
