Have you heard about Invitrogen's ThioFusion(tm) Expression System? The
system uses a vector of theirs to attach thioredoxin to a protein (presumably
whatever size peptide you decide to clone in). Once this is done, the
affinity column is relatively straightforward. The plasmid also contains an
enterokinase cleavage site, which will allow the thioredoxin to be released
from the desired native protein.
